Holly Hunter was born in Conyers, Georgia, the youngest of six children. Her first acting role was as Helen Keller in a fifth-grade play. She performed in high school productions of Oklahoma!, Man of La Mancha, and Fiddler on the Roof. She earned a drama degree from Carnegie Mellon University in Pittsburgh and performed in local theater before moving to New York City, where she roomed with Frances McDormand. A chance encounter with playwright Beth Henley in an elevator led to Hunter being cast in Henley's plays Crimes of the Heart and The Miss Firecracker Contest.

Hunter made her film debut in the 1981 slasher film The Burning. After moving to Los Angeles in 1982, she appeared in TV movies and supporting roles. She starred in the Coen brothers' Raising Arizona (1987) and received an Academy Award nomination for Broadcast News (1987). She won two Primetime Emmy Awards for Roe vs. Wade (1989) and The Positively True Adventures of the Alleged Texas Cheerleader-Murdering Mom (1993). In 1993, she won the Academy Award for Best Actress for The Piano and was also nominated for The Firm. Later nominations came for Thirteen (2003).

Hunter has been married to cinematographer Janusz Kamiński from 1995 to 2001. Since 2001, she has been in a relationship with British actor Gordon MacDonald, with whom she has twin sons born in January 2006. She is unable to hear with her left ear due to a childhood case of the mumps, which sometimes requires script alterations. She is irreligious.
